Procedure of HbA1C Test

HbA1C Test
HbA1C:   A form of glycated haemoglobin. It means glucose chemically attaches to heamoglobin without help of enzyme.
Normal Hb
                  1. HbA1:  97 %
                  2. HbA2:  2.5 %
                  3. HbF  :   0.5 %
Normal distribution of HbA1:
1. HbA1a
2. HbA1b
3. HbA1c —-80%
Specimen:
1. EDTA blood / cp bottle
Patient Preparation:
1. No preparation required (not affected by food, exercise and time of collection.)
Method:
1. HPLC (High performance liquid chromatography )-  Biorad D-50
2. Capillary electrophoresis
What are the factors effecting HbA1C:
1. Blood glucose level
2. RBC life spam
Normal Range:-  4.2% – 6.4%
Indication:
Diabetic patient:
a. To asses blood glucose level of previous 8-10 weeks(2-2.5 months)
b. Diagnostic for diabetic
c. To asses risk of diabetic complacent.

Quality Control: Quality control may be defined as the study source of errors which are the responsibility of the clinical laboratory and the process used to recognized and minimize them.
  
Types of Quality Control: 
1. Internal quality control
2. External quality control

Ans : 1.Accuracy : Ability of an analytical method to detect the test result, close to true value.
2. Precision: Ability of an analytical method to give the same result. If repeatedly in the same specimen for the same analyze.
3. Sensitivity: Ability of an analytical method to detect the smallest test quantity of substance than can be differentiated from zero.
4. Specificity: Ability of an analytical method to detect the substance that is specifically asked for.
